摘要 目的研究鬼针草提取物(BPE)对Hela细胞及A549细胞体外增殖的抑制作用及对细胞周期的影响。方法常规MTT法分析BPE对Hela细胞及A549细胞体外增殖的抑制作用并计算IC50;Hochest33342染色分析10μg/mLBPE对肿瘤细胞的凋亡诱导作用;流式细胞术分析BPE对肿瘤细胞周期的影响。结果BPE能抑制Hela细胞及A549细胞的体外增殖并呈剂量依赖性,IC50分别为10.34μg/mL及8.31μg/mL;Hochest染色证实BPE能诱导肿瘤细胞凋亡;流式细胞术分析证实BPE能将肿瘤细胞阻滞在G0/G1期。结论鬼针草能使细胞停滞于G0/G1期,并能诱导肿瘤细胞凋亡从而抑制肿瘤细胞的增殖。 Objective To investigate the effects of Bidens pilosa Linn. extract (BPE) on proliferation and cell cycle of Hela and A549 cells.Methods MTT was used to study the inhibition effect of BPE on the growth of Hela and A549 cells,based on which IC50 values were determined.The effect of BPE (10 μg/mL) on the apoptosis of Hela and A549 cells was tested by Hochest33342 staining and the percentage of apoptotic cells and changes in cell cycle were detected by flow cytometry.Results BPE inhibited the proliferation of Hela and A549 cells in vitro in a dose-dependent manner,with IC50 values as 10.34 and 8.31 μg/mL,respectively. BPE was testified to induce apoptosis of Hela and A549 cells and to block these cells in G0/G1 phase by Hochest staining and flow cytometry.Conclusion Hela and A549 cells were arrested in G0/G1 phase by BPE,which induced the increase of apoptotic cells and the decrease of proliferating cells.
