proBDNF对大鼠后足切割疼痛的影响 被引量:1

The Effects of proBDNF on Cutting Pain in Rats with the Hind Paw Incision

摘要 目的研究大鼠后足切割后proBDNF在脊髓和背根神经节的表达及注射proBDNF后大鼠的行为学变化。方法①16只大鼠随机分为切割组和对照组,切割组建立后足切割模型,取2组大鼠的脊髓和背根神经节行免疫荧光实验;②48只大鼠随机分为6组,每组8只,proBDNF腹腔注射大(Ⅰ组)、中(Ⅱ组)、小剂量组(Ⅲ组),鞘内注射组(Ⅳ组),足底注射组(Ⅴ组)和对照组(Ⅵ组)。各组大鼠建立后足切割模型,均行缩足阈值测定。结果①切割后1h大鼠背根神经节proBDNF的表达增强,脊髓proBDNF的表达没有明显变化;②Ⅰ组、Ⅱ组大鼠切割后大鼠右后足的机械痛阈明显高于Ⅲ组和Ⅵ组;Ⅳ组和Ⅴ组大鼠切割后大鼠右后足的机械痛阈与Ⅵ组相比无统计学意义。结论proBDNF在转运至脊髓之前已经转变为成体BDNF或者与其受体结合,腹腔注射proBDNF具有明显的止痛效果,并且其止痛作用有封顶效应。 Objective To study proBDNF's expression in the spinal cord and dorsal root ganglia after rat's hind paw incision and the changes of behavior after proBDNF injection.Methods ①16 rats were randomly divided into cutting group and control group. cutting model were set up in rats of cutting group.Immunofluorescence test detected the spinal cord and dorsal root ganglia from two groups of rats.②48 rats were randomly divided into proBDNF intraperitoneal injection of large(groupⅠ),medium(groupⅡ) and small dose group(groupⅢ),intrathecal injection group(groupⅣ),plantar injection group (groupⅤ)and control group(groupⅥ).All rats'hind-paw with drawal thresholds were measured after the establishment of cutting model.Results ①proBDNF's expression was enhanced in dorsal root ganglion 1 hour after incision and its expression in spinal cord had no significant change.②The rats'hind-paw withdrawal thresholds of Group Ⅰ,Ⅱ are higher than those of group Ⅲ and Ⅵ;The rats'hind-paw withdrawal thresholds of group Ⅳ,Ⅴ have no statistical significance compared with the groupⅥ.Conclusion proBDNF had become mature BDNF or combined with its receptor in prior to transportation to the spinal cord. Intraperitoneal injection of proBDNF had obvious analgesic effect,and the role of its analgesia had ceiling effect.
