摘要
目的:为了观察辐射诱导胶质瘤细胞凋亡的特征,进一步探讨辐射诱导肿瘤细胞凋亡与辐射剂量、剂量率和照射方式之间的关系。方法:将SWO细胞分为对照组、不同剂量组、不同剂量率组、单次照射组和分次照射组,以医用直线加速器X射线照射。采用光镜、电镜及DNA琼脂糖凝胶电泳法观察X射线诱导星形细胞瘤细胞系凋亡的特征。结果:(1)低剂量、低剂量率和分次照射组(10Gy)诱导出典型细胞凋亡的特征(如凋亡小体,DNA梯状带等)。(2)低剂量诱导肿瘤细胞凋亡,高剂量导致肿瘤细胞坏死。(3)凋亡出现时间因组织细胞类型的不同而各有差异。(4)X射线诱导肿瘤细胞的凋亡以分裂期凋亡为主。结论:(1)X射线诱导SWO细胞系的凋亡与剂量、剂量率和照射方式有密切关系,(2)本研究结果将为临床放射治疗脑胶质瘤的研究提供一定的新理论依据。
Purpose: To investigate the mechanism of radiation-induced apoptosis, the relationship between radiation-induced apoptosis and different radiation dose, dose-rate, radiation mode. Materials and Methods: SWO-38 cell lines from human astrocytoma were divided into control group and different radiation groups. Optical microscope, electron microscope and DNA agarose gel electrophoresis have been employed to observe the distinct biochemical and morphological features of the radiation-induced apoptosis in SWO cell lines after X-ray radiation by medical LINAC(linear accelerator) Results:(1)The classic morphological features of apoptosis (e.g. formation of apoptotic bodies) and the DNA fragmentation ladders on agarose electrophoresis were seen in the low dose's low dose-rate's and fractionation group.(2) Apoptosis was induced by low dose X-ray. Necrosis was caused by high dose X-ray.(3) The appearing time of apoptosis was variable in different tissues or cells.(4)Apoptosis induced by X-ray was almost mitosis death. Conclusion:(1)Apoptosis induced by X-ray in human astrocytoma cell lines was closely related to the dose, dose-rate and radiation mode.(2) These results provide some new theoretical basis for the clinical radiation treatment and research with glioma.
