大鼠三叉神经节及尾侧亚核内降钙素基因相关肽在面口部伤害性刺激时的动态变化 被引量:10

摘要 为了探讨降钙素基因相关肽(CGRP)在面口部伤害性刺激条件下合成、运输和释放的规律,将福尔马林注入大鼠一侧口周皮下,分别于刺激后不同时间取材,用原位杂交和免疫组织化学方法观察CGRPmRNA及CGRP样阳性结构(CGRP-LI)在三叉神经节(TG)和三叉神经尾侧亚核(Vc)内的动态变化。原位杂交结果显示:TG内表达CGRP的两种亚型α-CGRP和β-CGRPmRNAs神经元的数量分别于福尔马林刺激后2h和4h有明显增加,8h达到高峰,然后渐下降,至48h后恢复至正常。免疫组织化学结果显示:Vc浅层CGRP-LI的含量在刺激后1h内明显减少,2h后渐增,12h达到高峰,24h后趋向正常。TG内CGRP-LI神经元的数量在刺激后1h内也明显减少,此后逐渐回升至正常并有轻度增加,但与正常间无明显差异。 To detect the changes of the systhesis,transportation and release of the calcitonin gene re lated peptide(CGRP) mRNAs and CGRP like immunoreactivity(CGRP LI) in the caudal spinal trigeminal nucleus(Vc) and trigeminal ganglion(TG)subsequent to subcutaneous injection of formalin into oroficial region of the rat, in situ hybridization histochemical and immunohistochemical methods were used in the present study.The results showed that significant increases of αCGRP and βCGRP mRNAs expression in the ipsilateral TG were observed 2h and 4h after formalin injection,respectively,reached the peak at 8h and returned to normal level 48h later.CGRP LI in the superficial laminae of the Vc was significantly decreased ipsilaterally from 0 5h to 1h after injection,then increased gradully from 2h to 12h.After 24h,there were no remarkable differences in the level of CGRP LI between two sides of the Vc.The number of CGRP like positive neurons in the ipsilateral TG decreased within 1h after stimulation,then showed a slight increase from 2h to 12h.
