目的总结提高免疫组织化学切片质量的经验体会。方法在免疫组织化学实验过程中,通过提高组织固定质量、制备高质量的载玻片、选择最佳的蜡块、制备高质量的切片、合适的抗原修复、严谨进行染色操作、选择合适的检测系统、设立有效的实验对照等措施提高免疫组织化学切片质量。结果阳性细胞呈棕黄色-棕裼色颗粒,阳性颗粒定位于抗原所在的部位上,背景清晰。结论免疫组织化学染色技术具有特异性强、灵敏度高、定位准确等优点,严格实验操作,不断优化实验方法,提高其染色质量,在病理诊断与科研工作中具有重要的作用。

Objective To sum up the experience in improving quality of sections with immunohistoehemical staining. Methods The quality of tissue fixation, preparation of high - quality glass slides, choosing best wax blocks, high - quality slice preparation, appropriate antigen retrieval, critical dyeing operation, selection of appropriate detection system and effectively experimental control of immunohistochemical process were enhanced in order to raise the quality of staining. Results Positive cells had brown - yellow to brown color Ti particles, and these particles were located in antigenic site under clear background. Conclusion Immunohistochemieal staining technique has following advantages, such as strong specificity, high sensitivity and accurate positioning. Strictly mastering its operational procedure is important in optimizing and improving the quality of staining, and it will play an important role in pathological diagnosis and research work.
