摘要 目的:建立同时测定大鼠血浆中盐酸小檗碱、大黄素、大黄酚和大黄素甲醚含量的方法。方法:采用高效液相色谱串联质谱法,色谱柱为Shim Pack VP-ODS C_(18)色谱柱(150 mm×2.0 mm,5μm),柱温30℃,流动相为乙腈:0.5%甲酸水溶液(80:20,氨水调pH=3.5),流速0.2ml·min^(-1)。结果:盐酸小檗碱、大黄素、大黄酚、大黄素甲醚的最低检测限范围为1.0~1.2ng·ml^(-1),定量下限范围为5.0~11.5 ng·ml^(-1),日内、日间RSD<2.98%。结论:该法准确、快速,可用于上述4种成分的药动学研究。 Objective : To Establish a LC-ESI-MS method to quantify the concentrations of berberine hydroehloride, emodin, chryr sophanol and physcion in rat plasma after oral administration of Sanhuang tablet. Method: Liquid chromatography coupled with Mass spectrometry, with Shim Pack VP-ODS Cls analytical column (150 mm × 2.0 mm, 5 μm)was used, oven temperature was 30%1 and the mobile phase was a mixture of acetonitrile and 0.5% formic acid solution ( adjust pH to 3.5 with ammonia) , flow rate was 0.2 ml · min^ - 1 . Result : The limit of determinations ( LODs ) of the three constituents were ranged from 1.0 to 1.2 ng · ml^ - 1 , the lower limit of quantitations (LLOQs) were ranged from 5.0 - 11.5 ng · ml ^-1 , RSDs of intra-day and inter-day precision were less than 5%. Conclusion: The method could be used for the fast and accurate determination of the four components in rat plasma, and it was successfully applied for the investigations of their pharmaeokinetic properties.
