摘要
目的:建立测定罗汉果中多糖、总糖及罗汉果苷V含量的方法,并测定其不同部位的含量。方法:采用苯酚-硫酸比色法测定多糖及总糖含量,测定波长为487nm;用眦测定罗汉果苷V含量,PhenomenexC_(18)柱,乙腈-水(22:78)为流动相,流速1.0 ml·min^(-1),检测波长208 nm。结果:无水葡萄糖在32.0~192.2μg,r=0.999 8,罗汉果苷V在0.81~12.1μg范围内具有良好的线性关系(r=0.999 9),平均回收率分别为100.7%和102.0%,RSD为2.33%和1.03%(n=6);其含量均以果肉最高而种子较低。结论:方法灵敏、准确,重复性好,适用于该药材的质量评价。
Objective: To establish a methods for determination the contents of polysaccharides, saccharides and mogroside V in Fructus Momordicae from various sample and part. Method: For poiysaccharides and saccharides, the phenol sulferic acid colorimetric method was developed and some factors were also optimized,the detection wavelength was 487 nm, the mogroside V content of Fruetus Momordicae was determind by HPLC. Result: The calibration curves of glucose anhydrous and mogroside V were linear within the range of 32.0-192.2 pLg ( r = 0. 999 9 ), 0.81-12.1 p.g ( r = 0.999 9 ), the average recoveries ( n = 6 ) were 100.7 %, 102.0%. with RSD were 2.33% , 1.03% respectively, their contents were the highest in flesh and peel, Lowerest in seed. Conclusion: The method is sensitive, accurate and repeatable. It can be used for quality contrnl of Fructus Momordicae.
