摘要
目的研究Th17细胞相关细胞因子IL-23和IL-17在寻常型天疱疮(PV)发病机制中的作用。方法采用免疫荧光染色法分析PV患者皮损组织和健康人皮肤组织中Th17细胞相关因子IL-17和IL-23的表达及CD163^+巨噬细胞的分布。结果 IL-23^+细胞的染色与CD163^+巨噬细胞的染色重叠。PV患者真皮中IL-23^+ CD163^+细胞和IL-17^+细胞在每个视野(×200)中的平均数分别为80.17±5.42和48.17±8.03,而正常人真皮中分别为29.50±4.45和9.50±3.35,两组差异均有统计学意义(P<0.01)。真皮层中IL-23^+细胞数和IL-17^+细胞数存在相关性(r=0.945,P<0.001)。结论 CD163^+巨噬细胞是IL-23重要分泌源;IL-23和IL-17在PV患者皮损组织中高表达,IL-23/IL-17细胞因子轴在PV发病机制中可能起着非常重要的作用。
Objective To investigate the role of Thl7 cell associated cytokines of IL-23 and IL-17 in the pathogenesis of pemphigus vulgaris. Methods Immunofluorescence staining was performed to analyse the distribution of CD163~+ macrophages and expression of IL-23 and IL-17 in pemphigus vulgaris lesions and healthy human skin tissues. Results The staining of IL-23~+ cells is overlapped with that of CD163~+ macrophages. The mean numbers of IL-23~+ CD163~+ cells and IL-17~+ cells were 80.17 ± 5. 42 and 48. 17 ± 8. 03 in each field of view (×200) in dennis of pemphigus vulgaris, while those in healthy human dermis were 29. 50 ± 4. 45 and 9. 50 ± 3. 35, with significant differences between dennis of pemphigus vulgaris and healthy human dermis (P 〈0.01). The number of IL23~+ cells was related to that of IL-17~+ cells in dermis (r =0.945, P〈0.001). Conclusion IL-23 is mainly secreted by CD163~+ macrophages. IL-23 and IL-17 are overexpressed in pemphigus vulgaris lesions. IL-23/IL-17 cytokine axis may play an important role in the pathogenesis of pemphigus vulgaris.
