摘要 目的:探讨神经生长因子(NGF)和碱性成纤维细胞生长因子(bFGF)诱导大鼠乳鼠肌源性干细胞(MDSCs)分化为神经样细胞的可行性.方法:取SD大鼠乳鼠骨骼肌,差速贴壁至第5代时培养液中加入诱导培养基培养,行神经元特异性烯醇化酶(NSE)免疫细胞化学鉴定和RT-PCR分析.结果:分化后细胞NSE染色阳性,RT-PCR结果显示,诱导后MDSCs样品中检测到NSE、GFAP特异性条带.结论:在一定条件下NGF和bFGF联合诱导法可以诱导MDSCs分化为神经样细胞. Objective: To explore the feasibility of inducing muscle-derived stem cells into neuron-like cells in vitro. Methods: The skeletal muscle stem cells were isolated with mixed digestive enzyme, cultured and purified clonally in vitro. The 5th passage ceils were incubated in serum free medium including basic fibroblast growth factor (bFGF) and nerve growth factor(NGF). Morphological change was detected and phenotype was analyzed by immunoeytochemistry and RT-PCR during incubation period. Results: After induction, the cells became non-adherent aggregates as neuron-like cells. In differentiation condition, they were like neurons and glial cells and expressed imrnuno-positive for NSE. The result analyzed by RT-PCR was coincident with that by immunocytochemistry. Conclusion: Muscle-derived stem cells of rats can be induced into neuronlike cells in vitro.
