摘要
目的探讨经典神经递质去甲肾上腺素(norepinephrine,NE)对人胰腺癌细胞株MiaPaCa-2侵袭能力的影响及其作用机制。方法取人胰腺癌细胞株MiaPaCa-2为研究对象,实验分为空白对照组、0.1μmol/L NE干预组、1μmol/L NE干预组和10μmol/L NE干预组。干预48 h后,采用Transwell侵袭实验检测各组中MiaPaCa-2细胞侵袭能力的变化;RT-PCR法检测细胞中MMP-2、MMP-9和VEGF mRNA的表达;免疫细胞化学法检测细胞中MMP-2、MMP-9和VEGF蛋白的表达。结果1μmol/L NE干预组和10μmol/L NE干预组MiaPaCa-2细胞的侵袭能力均高于空白对照组(P<0.01);1μmol/L NE干预组和10μmol/L NE干预组MiaPaCa-2细胞中MMP-2、MMP-9和VEGF mRNA和蛋白表达水平均高于空白对照组(P<0.05)。结论NE可以通过上调MMP-2、MMP-9和VEGF的表达从而促进胰腺癌细胞发生远处侵袭转移。
Objective To investigate the effect of norepinephrine(NE) on the invasiveness of pancreatic cancer cell line MiaPaCa-2 and its action mechanism.Methods The pancreatic cancer cell line MiaPaCa-2 was used in the research.The cells were randomly divided into control group,0.1μmol/L NE intervention group,1μmol/L NE intervention group and 10μmol/L NE intervention group.After intervention for 48h,Transwell invasiveness test was given to examine the changes of invasive ability of MiaPaCa-2.The expressions of MMP-2 mRNA,MMP-9 mRNA and VEGF mRNA were measured by semi-quantitative RT-PCR;the levels of MMP-2,MMP-9 and VEGF proteins were measured by immunocytochemical method.Results The invasive ability of MiaPaCa-2 in 1μmol/L NE intervention group and 10μmol/L NE intervention group was significantly higher than that in control group(P〈0.01).The levels of MMP-2,MMP-9 and VEGF mRNA and protein expression of MiaPaCa-2 in 1μmol/L NE intervention group and 10μmol/L NE intervention group were higher than those in control group(P〈0.05).Conclusion NE can promote the invasiveness of MiaPaCa-2 through upregulating the expressions of MMP-2,MMP-9 and VEGF.
