摘要
目的观察单侧黑质纹状体通路损毁对大鼠LDT神经元电活动的影响。方法采用玻璃微电极细胞外记录方法观察和分析正常大鼠及PD大鼠LDT神经元放电频率和放电形式的变化。结果对照组和实验组大鼠LDT神经元的平均放电频率分别是(8.24±3.37)Hz(n=40)和(12.69±6.28)Hz(n=52)。实验组大鼠LDT神经元的放电频率显著高于对照组(P<0.05)。对照组大鼠LDT神经元规则、不规则,爆发式放电分别占7.5%,77.5%,和15%,实验组大鼠规则、不规则,爆发式放电的神经元分别占1.9%,82.7%和15.4%,两组大鼠之间LDT神经元的放电形式未见显著性差异(P>0.05)。结论帕金森病大鼠LDT神经元的放电频率显著增高(P<0.05),放电形式无显著变化,提示PD状态下,LDT神经元兴奋性活动占主导地位。
Objective To investigate the effects of the destroyed unilateral nigroatrital pathway on neuronal activity of the rat laterodorsal tegmental nucleus(LDT).Methods The changes in the firing rate and firing pattern of the LDT neurons were observed by extracellular recording method in control and Parkinson's disease(PD) rats.Results The results showed that the mean firing rate of LDT in control and PD rats was(8.24±3.37)Hz(n=40) and(12.69±6.28)Hz(n=52),respectively.The mean firing rate of PD rats was significantly increased when compared with that of the control rats(P〈0.05).In the control rats,the regular,irregular and bursting firing neurons recorded were 7.5%,77.5% and 15%,respectively while those in PD rats were 1.9%,82.7% and 15.4%,respectively.There was no significant difference in the firing pattern between PD and control rats(P〉0.05).Conclusion The firing rate of neurons in LDT was increased in PD rats;however,the firing pattern did not change significantly.It indicates that the exciting activity of LDT plays a dominant role in PD rats.
