摘要
目的:探讨HaCaT细胞在HSV-II刺激前后角质细胞生长因子(keratinocyte growth factor,KGF)和转化生长因子α(Transforming growth factor-α,TGF-α)的分泌变化及意义。方法:以VERO细胞进行HSV-II扩增;采用RT-PCR和荧光实时定量PCR检测HSV-II感染HaCaT细胞前及感染后24h、48h、72hKGF和TGF-α的分泌量。结果:HaCaT细胞可自分泌KGF和TGF-α;感染HSV-II后,KGF明显升高,而TGF-α的分泌与对照组相比差异无统计学意义。结论:HaCaT细胞可以自发分泌KGF和TGF-α;KGF可能参与HSV-II感染角质形成细胞(keratinocyte,KC)后皮肤的修复过程;而TGF-α在此过程中不起主要作用。
Objective:To investigate the influence of HSV-2 on KGF and TGF-α expression levels in HaCaT cells, and to explore the potential significance of KGF and TGF-αin HSV infection. Methods:HSV-2 was amplified in VERO cells. HaCaT cells were infected with HSV-2, and were harvested 24 h, 48 h and 72 h after infection. Expression levels of KGF and TGF-α were detected by Real-time PCR. Results: ExPression of KGF and TGF-α can be detected in normal HaCaT cells. After HSV-2 infection, expression level of KGF was higher in the HSV-2 infected groups than that in the control ( P 〈 0.05 ), while expression level of TGF-α did not change significantly. Conclusion: HaCaT cells express KGF and TGF-α. KGF may play a positive role on the regeneration of epidermis after HSV-2 infection, while TGF-α may not have the same effect.
