芽孢杆菌碱性蛋白酶的沉淀分离特性和酶学性质研究

摘要 土壤中筛选获得一株产碱性蛋白酶的芽孢杆菌。该蛋白酶发酵16h达到产酶高峰,蛋白酶热稳定性较好,50℃条件下处理50min,酶活性残留86%,最适pH值7.7,丙酮、PEG等可有效沉淀蛋白酶,(NH4)2SO4在5℃,pH9.0条件下可沉淀分离蛋白酶,回收率达95%。 A strain Bacillus sp.,of antioxidant alkaline protease,was screened.Its enzymatic activity is maximum after 16h fermentation.The enzyme was stable at 50℃ for 50min,retaining more than 86% of its activity.Optimum pH for enzyme activity was pH7.7.Protease was effectively separated by acetone,PEG.The specific activity was 95% yield when treated by (NH4)2SO4 at pH9.0 and 5℃.
