摘要
目的建立以蒸发光散射检测器(ELSD)和二极管阵列检测器(DAD)测定威灵仙药材中齐墩果酸和常春藤皂苷元的高效液相色谱分析方法。方法色谱柱:Merck Lichrocart C18柱(4.0 mm×250 mm,5μm);流动相:甲醇-水-冰醋酸-三乙胺(体积比87∶13∶0.04∶0.02);流速:1.0 mL.min-1;柱温:30℃;DAD检测波长为210 nm;ELSD蒸发管温度为80℃,气体压力为172.375 kPa。结果常春藤皂苷元、齐墩果酸线性范围分别为1.43-14.3μg和2.1-21.0μg;常春藤皂苷元和齐墩果酸在以DAD检测时的加样回收率分别为98.02%、98.94%,RSD分别为2.61%、2.34%;在以ELSD检测时的加样回收率分别为98.72%、100.03%,RSD分别为2.51%、2.22%。结论本方法简便、重现性好、专属性强,为完善威灵仙药材质量控制标准提供了科学依据。
Objective To determine the content of oleanolic acid and hederagenin from Radix Rhizoma Clematidis by RP-HPLC-ELSD-DAD at the same time. Methods A Merck Lichrocart C18 column(4.0 mm×250 mm,5 μm) was used.The mobile phase was consisted of methanol-water-acetic acid-triethyl amine(87∶13∶0.04∶0.02) with a flow-rate of 1.0 mL·min^-1,and the column temperature was 30℃.The DAD detection wavelength was 210 nm;The ELSD drift-tube temperature was 80℃,and gas pressure was 172.375 kPa.Results The linear ranges of hederagenin and oleanolic acid were 1.43-14.3 μg and 2.1-21.0 μg respectively.The average recovery of hederagenin and oleanolic acid were 98.02%(RSD=2.61%),98.94%(RSD=2.34%) determined by DAD,and 98.72%(RSD=2.51%),100.03%(RSD=2.22%)determined by ELSD.Conclusion It's a reliable and accurate method for the quality control of R.Clematidis.
