摘要
目的探讨ABCG2在人脑胶质瘤组织中的表达及其与胶质瘤分级和Cyclin D2表达的关系。方法实时荧光定量PCR法检测ABCG2在52例不同病理级别胶质瘤及8例正常脑组织中的mRNA表达;免疫组织化学SABC法检测ABCG2和Cyclin D2的蛋白表达;SPSS统计分析其表达的相关性。结果ABCG2在脑胶质瘤中的mRNA表达与正常脑组织相比存在过表达(P<0.05),且随着病理级别的升高而增加(P<0.05);免疫组化检测显示ABCG2在52例胶质瘤中的阳性表达率为32.7%(17/52),与病理级别具有明显相关性(χ2=4.62,P<0.05),呈亲血管分布;ABCG2的表达与Cyclin D2的表达具有一定的相关性(P<0.05)。结论ABCG2在人脑胶质瘤中的表达随着病理级别的升高而逐渐增加,且与Cyclin D2的表达呈正相关,表明以ABCG2为标志物的脑肿瘤干细胞具有较高的增殖能力。
Objective To investigate the expression and significance of ABCG2 and its relationship with WHO pathological grade and Cyclin D2 in human gliomas.Methods Fifty-two cases of gliomas with different WHO grades and 8 cases of normal brain tissues were detected for the mRNA expression of ABCG2 by using real-time quantitative PCR.The protein expression of ABCG2 and Cyclin D2 was assayed by immunohistochemistry.The correlation was analyzed by SPSS 12.0 software.Results The mRNA of ABCG2 was overexpressed in gliomas as compared with that in normal brain tissues,and up-regulated with the increase of pathologic degree(P0.05).The positive protein expression rate of ABCG2 in 52 cases of gliomas was 32.7%.The positive rate was significantly higher in grade Ⅲ-Ⅳ than that in grade Ⅰ-Ⅱ(χ2 =4.62,P0.05).The expression level of Cyclin D2 was obviously related with the expression of ABCG2(P0.05).Conclusion The expression level of ABCG2 was obviously related with glioma pathological grade and the expression of Cyclin D2.Brain tumor stem cells characterized by positive expression of ABCG2 may show heavy potency of malignant proliferation.
