摘要 目的分离培养新生SD大鼠神经干细胞,并对其生物学特性进行鉴定。方法采用无血清培养联合应用碱性成纤维细胞生长因子和表皮生长因子对大鼠海马神经干细胞分离培养,采用免疫荧光的方法检测神经干细胞的nestin抗原。结果从新生大鼠海马齿状回分离的细胞群可不断增殖形成细胞克隆球,并且呈nestin阳性表达,具有多向分化能力。结论分离培养的细胞是中枢神经系统的干细胞。 Objective To isolate and culture neural stem cells from SD newborn rats,and identify its biological properties. Methods This study adopted the serum -free culture combined with the basic fibroblast growth factor and epidermal growth factor to isolate and culture the rat hippocampal neural stem cells, using immunofluorescence method to detect the nestin antigen of neural stem cells. Results The results showed that the cells isolated from the hippocampal dentate gyrus could proliferate continuously,and form the cell clones with multi-directional differentiation, and show positive expression of nestin. Conclusion Those isolated and cultured cells are the stem cells of the central nervous system.
