替勃龙对阿尔茨海默病模型大鼠的学习记忆能力及神经元凋亡的影响

摘要 目的观察替勃龙(tibolone)对AD大鼠的学习记忆能力及神经元凋亡的影响。方法用立体定位技术对海马CA1区微量注射Aβ建立AD模型;Morris水迷宫检测大鼠记忆水平;TUNEL染色法检测海马凋亡。结果替勃龙可明显提高Aβ所致痴呆大鼠学习记忆水平,显著降低海马细胞凋亡率。结论替勃龙对AD大鼠的保护作用与降低海马细胞凋亡率有关。 Objective To explore the effect of tibolone on inhibiting apoptosis of hippocampus cells and learning and memory in Aβ-induced Alzheimer disease rat model. Methods The AD pathological model was made through injection of Aβ 25-35 in hippocampal CA1, and the learning and memory of rats were observed with Morris water maze; the apoptosis of hippocampal cells were detected by using TUNEL. Results Compared with the control group, tibolone was shown to improve learning and memory in rats with bilateral hippocampal lesions, and decrease apoptosis rate in hippocampal cells significantly. Conclusion These results suggest that the protective effects of tibolone on AD rats may be related to its ability to decrease the apoptosis rate of hippocampal cells.
