摘要
目的研究慢性复合应激对大鼠血清脂联素水平及肝脏脂联素受体-2(AdipoR2)表达的影响。方法将40只SD大鼠随机分为应激组和对照组,每组20只。应激组实施8周的慢性复合应激刺激,对照组在此期间正常饲养。在实验的第8周将大鼠断头处死,取血检测脂联素、血糖、胰岛素抵抗(IR)、糖化血红蛋白(HbA1C)、高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL—C)、甘油三酯、总胆固醇含量。结果①8周后,应激组与对照组旷场实验得分和24h尿皮质醇差异有统计学意义[(32.0±8.6)、(52.0±12.7)分,(5.8±0.4)、(5.3±0.1)ng/L,P〈0.05或〈0.01]。②8周后应激组与对照组脂联素、总胆固醇,甘油三酯、HDL—C、1nIR、HbA,C差异有统计学意义[(4.97±0.57)、(5.63±0.49)mg/L,(1.44±0.24)、(1.07±0.23)mmo]/L,(0.47±0.09)、(0.39±0.05)mmol/L,(0.76±0.17)、(0.89±0.15)mmol/L,(1.89±0.46)、(1.41±0.32),(7.84±1.72)%、(6.08±1.56)%,t值分另q为-3.927、4.978、3.475、-2.564、3.831、3.390,P〈0.05或〈0.01]。③8周后应激组和对照组大鼠肝脏AdipoR2 mRNA相对表达分别为0.67±0.04、1.00±0.11,蛋白相对表达分别为0.73±0.02、1.00±0.04,差异有统计学意义(t值分别为-12.609、-27.000,P均〈0.01)。脂联素与HbA1C、LDL-C、IR显著负相关(r值分别为-0.26、-0.88、-0.37,P〈0.05或P〈0.01),与HDL—C显著正相关(r=0.78,P〈0.01)。结论慢性复合应激使大鼠血清脂联素水平和肝脏AdipoR2表达降低。
Objective To explore the effect of chronic compound stress on serum adiponectin level and AdipoR2 expression in rats. Methods Forty male Sprague-Dawley rats were randomly divided into two groups:the stress group underwent chronic compound stress and the control group was fed normally. After 8 weeks of stress or feeding, the serum adiponectin, blood glucose,insulin resistance and HbA, C, HDL-C, LDL-C, triglyceride and total cholesterol were measured. Results ①After 8 weeks of stress, the score of open-field behaviors and 24-h urocortisol were significantly different between the stress and control groups(32. 0 ±8.6 vs. 52.0 ± 12. 7,5.8 ±0. 4 vs. (5.3 ± 0. 1 ) ng/L, repectively ; P 〈 0. 05 or 0. 01 ) ;②After 8 weeks of stress, the adiponectin, total cholesterol, triglyceride, HDL-C, lnlR and HbA1C were significantly different between the two groups ( P 〈 0. 05 or 0. 01 ) ; ③After 8 weeks of stress, there were significant difference on the expression of AdipoR2 mRNA and protein between the two groups (P 〈 0. 01 ), with the expression of AdipoR2 mRNA of 0. 67 ±0. 04 and 1.00 ± 0. 11 and the expression of protein of 0. 73 ± 0. 02 and 1.00 ± 0. 04 in the stress and control group, respectively. Adiponectin was negatively related with HbA1C, LDL-C and IR (r = -0. 26, -0. 88 and -0. 37,P 〈 0.05 or 0. 01 ), and positively related with HDL-C (r = 0. 78 ,P 〈 0. 01 ). Conclusions In rats, chronic compound stress decreases the level of serum adiponectin and the expression of AdipoR2 in liver.
