不同剂量黄连素对心肌缺血大鼠的抗氧化损伤作用 被引量:7

摘要 目的研究不同剂量黄连素(盐酸小檗碱)对实验性心肌缺血大鼠抗氧化损伤作用。方法40只雄性Wistar大鼠随机分为正常对照组、假手术组、模型组、黄连素低剂量组[50mg/(kg.d)]、黄连素高剂量组[150mg/(kg.d)],每组8只,除正常对照组和假手术组外,均采用心脏冠脉结扎法建立大鼠心肌缺血模型,各组灌胃治疗7d。心脏取材后观察心脏组织病理变化,检测组织中超氧化物歧化酶(SOD)活性和丙二醛(MDA)含量改变。结果与正常对照组比较,模型组SOD活性和MDA含量显著增加(P<0.05);与模型组比较,黄连素高、低剂量组MDA含量均显著降低(P<0.05),黄连素高剂量组降低更为显著(P<0.01);黄连素低剂量组SOD活性显著增加,黄连素高剂量组无显著增加。结论黄连素可减轻心肌缺血大鼠心肌的氧化损伤而对心肌起到保护作用,其作用与浓度有一定依赖关系,黄连素高、低剂量组的作用机制和途径可能不同。 Objective To investigate the effects of berberine on the oxidative damage of myocardial ischemia in rat heart. Methods 40 male Wister rats were randomly divided into 5 groups:control group, sham group,model group, low-dose[50 mg/(kg·d)]group and high-dose[150 mg/(kg·d)]group. Except rat in control group, myocardial ischemia models of rat were established by cardiac coronary ligation. Berberine was given by mouth in low-dose group and high-dose group for 7 days. 7 days later, the animal were anesthetized with pentobarbital sodium (50 mg/kg, ip), the pathological changes of heart tissues were observed, MDA and SOD were determined by using kit. Results Compared with control group, SOD activity and MDA content in heart tissues were increased in model group (P〈0.05). Compared with model group MDA in low-dose group and high-dose group was decreased (P〈0.05, P〈0.01). SOD activity in lose-dose group was increased compared with model group (P〈0.05).Conclusion Berberine can resist oxidative damage of myocardial ischemia and protect heart tissue.
