摘要
目的:探讨MCI-186对VD大鼠海马组织Caspase-3的影响。方法:采取双侧颈总动脉永久性结扎的方法制备VD大鼠模型,分MCI-186大、小剂量治疗组、模型组、假手术组和正常对照组。通过Morris水迷宫实验检测大鼠学习记忆的改变。采用免疫组化技术检测凋亡调控基因Caspase-3蛋白的表达。结果:MCI-186能改善VD大鼠的学习记忆能力,并可明显抑制神经细胞凋亡,对抑制Caspase-3的表达均有显著作用。结论:证明MCI-186对慢性缺血致VD大鼠具有治疗作用,其作用机制与抑制神经细胞凋亡的表达有关。
Objective:To investigate the effect of MCI--186 on VD rat hippocampus Caspase-3. Methods: The VD rat models were prepared by a permanent ligation of the bilateral common carotid artery. There were four groups: large- and low- dose treatment group, model group, sham- operated group and normal control group. The changes in learning and memory of rats were detected by using Morris water maze test. The apoptosis regulatory gene Caspase- 3 protein expression was detected by immunohistochemical technique. Results: MCI-186 could improve the learning and memory of VD rats and could significantly inhibit both the neuronal apoptosis and the of Caspase--3 expression. Conclusion :It is suggested that MCI-186 has a therapeutic effect on VD rats induced by chronic ischemia, and its mechanism is correlated with the inhibition on the expression of neuronal apoptosis.
