摘要
为建立更有效的乙肝疫苗,通过将所构建的HBVRNA疫苗和DNA疫苗分别转染BHK-21细胞,并比较其在细胞中表达情况及细胞凋亡现象。以pSFV为基础构建的疫苗载体的表达效率高于以pcDNA3.1为基础构建的疫苗载体,pSFV为基础构建的疫苗载体出现细胞凋亡的现象。HBVRNA疫苗比DNA疫苗表达效果更好,安全性更高,更具有应用前景。
To establish a more effective hepatitis B vaccine, The HBV RNA vaccines and DNA vaccines were transfected into BHK-21 cells, and compare its expression in cells and cell apoptosis. The results show that the vector based on pSFV because BHK-21 cells apoptosis, and futher study indicated that the immune effect of RNA replicon vaccine is better than that of DNA vaccine. HBV RNA vaccine is better than the DNA vaccines, RNA vaccine is safer and have more prospects in gene therapy.
