摘要
为了更好地保护霍山石斛野生资源,变野生为家种,以霍山石斛试管丛生芽为试验材料,采用3因素3水平L(933)正交设计方案,考察BA、NAA、KT不同激素及浓度对霍山石斛试管丛生芽继代增殖的影响。结果表明:植物激素BA、NAA、KT对霍山石斛试管丛生芽的增殖均有一定影响,其中NAA及KT的影响更为明显,并通过SAS软件统计分析确定3种植物激素的最佳浓度配比分别为:BA1.5mg/L,NAA1.0mg/L,KT1.0mg/L。即霍山石斛试管丛生芽继代增殖的最佳培养基配方为:MS+BA1.5mg/L+NAA1.0mg/L+KT1.0mg/L+食糖30.0g/L+琼脂8.0g/L。
In order to protect the wild resources of Dendrobium huoshanense in better and change their home of wild species. In vitro shoots of Dendrobium huoshanense test material, using three factors, three level L9(33) orthogonal design to study BA, NAA, KT of different hormones and concentration on the propagation of adventitious buds of Oendrobium huoshanense subculture proliferation. The results showed that: plant hormones BA, NAA, KT right Dendrobium huoshanense proliferation of in vitro multiple shoot clumps had a certain impact, in which the impact of NAA and KT even more pronounced and, through the SAS statistical analysis software to determine the optimal concentration of three kinds of plant hormones ratio were as follows: BA use 1.5 mg/L, NAA use 1.0mg/L, KT selected 1.0 mg/L. Namely, the best medium formula of Dendrobium huoshanense subculture in vitro multiple shoot clumps were as follows: MS+BA 1.5 mg/L+NAA 1.0 mg/L+KT 1.0 mg/L+ Sugar 30.0g/L+ Agar 8.0 g/L.
