中国国内首次检测到犬细小病毒CPV-2c 被引量:44

The first detection of canine parvovirus type 2c in China

摘要 目的分析犬细小病毒(CPV)流行毒株的分子生物学特征,研究犬细小病毒遗传进化规律,了解CPV流行毒株现状。方法收集CPV阳性犬粪PCR扩增CPV-2c后进行RFLP分析,并进行病毒结构蛋白VP2基因的克隆、测序并进行序列分析。结果经RFLP分析及序列测定,2份样品均为CPV-2c阳性,序列分析显示CPV-06/09及CPV-07/09VP2序列与国外CPV-2c的基因序列同源性在99%以上。进化分析表明CPV-06/09及CPV-07/09在进化树中形成一个独立的分支,不同国家的CPV-2c存在一定的差异。结论通过RFLP分析及测序证实2份犬粪便样品为CPV-2c阳性,首次在我国检测到CPV-2c。系统进化树显示两分离株CPV-2c与国外CPV-2c有所差异,可能是传入我国的CPV-2c在向适应地区条件的方向进化,也可能是我国已有CPV变异。 Objective To ascertain the molecular biological characteristics of canine parvovirus(CPV)and analyze the current epidemiological state of CPV infection.Methods CPV-positive fecal samples from pet dogs were collected from veterinary hospitals and subjected to PCR-RFLP analysis.The VP2 gene of CPV-2c was cloned and sequenced;sequence analysis was carried out using DNAstar and MEGA 4.0.Results PCR-RFLP analysis and sequence analysis indicated that 2 out of the 12 samples were CPV-2c-positive.Sequences of CPV-06/09 and CPV-07/09 had homology of 99% or more with sequences of CPV-2c isolated outside China.A phylogenetic tree revealed differences in CPV-2c isolated in different countries,and CPV-06/09 and CPV-07/09 formed a separate cluster in the phylogenetic tree.Conclusion This study confirmed the existence of CPV-2c in China,and this is the first report of CPV-2c infection in China.Differences in CPV-2c isolated in different countries were attributed to local adaptation of CPV,and the genetic diversity of CPV isolated in different regions indicates that CPV is continuing to evolve.
