摘要
目的探讨骨桥蛋白(OPN)及IL-18在大鼠肾移植急性排斥反应中的变化规律及早期预测排斥反应的价值。方法采用实时荧光定量PCR检测移植肾OPN及IL-18 mRNA表达。ELISA法检测血清OPN及IL-18的浓度。结果移植肾组织中OPN mRNA在术后第三天表达显著上调,第五天达到高峰(106.79±8.34VS15.67±1.36,P〈0.05),第七天持续高表达。IL-18mRNA在第五天表达显著上调,达到高峰(76.00±5.47vs6.82±0.44,P〈0.05),第七天仍持续高表达,外周血中OPN及IL-18的浓度有相同的变化规律。对照组术后移植肾及外周血OPN及IL-18未见明显变化。结论OPN及IL-18均在肾移植急性排斥反应发生时出现表达上调,OPN在早期表达上调更显著;它们可以作为肾移植后急性排斥反应的早期检测指标,OPN可能是更理想的早期检测指标。
Objective Investigate the value of the OPN and IL- 18 expressions in the early diagnosis of acute renal allograft rejection in the rat. Methods Use real - time PCR to detect the expressions of OPN and IL- 18 mRNA in renal. Use enzyme - linked immunosorbent assay(ELISA)to detect the concentration of OPN and IL- 18 in blood. Results In the tissue, the expression of OPN mRNA up - regulated at day 3 and reached the high point at day 5 ( 106.79 ± 8.34 vs 15.67 ± 1.36, P 〈 0.05 ) .Meanwhile,the expression of IL- 18 mRNA up- regulated at day 5 and continue up regulated till the day 7(76.00 ±5.47 vs 6.82 ±0.44, P 〈 0.05).The expression of OPN and IL- 18 did not differ siginificentily change in the control group. Conclusion OPN and IL- 18 both up- regulated in acute renal allgraft transplantion in rats, and OPN up- regulates earlier; OPN and IL- 18 both could be sensitive factors in early diagnosis of acute rejection in rat transplantion, and OPN may be better.
