SD大鼠绝经模型建立的研究

The Study on the Building of SD Rats Models of Menopause

目的探讨利用大鼠卵巢切除术建立绝经模型及验证模型的关键技术问题,为大鼠绝经模型的建立提供经验。方法4月龄史-道(SD)雌性大鼠20只,10只采用背部脊柱两侧切口,切除卵巢,10只作为正常对照,利用吉姆萨染色法观察阴道脱落细胞,验证模型,并记录图像。采用酶联免疫法检测血清雌激素、卵泡刺激素(FSH)水平,比较两组大鼠血清性激素水平的变化。结果10只大鼠双侧卵巢切除完整,去势组与对照组相比雌激素水平显著下降(P<0.05),FSH水平差异无统计学意义(P>0.05),术后无伤口感染,1只切口裂开。结论该卵巢摘除方法具有切口小,操作简便,成功率高的特点,大鼠绝经模型激素水平标准的建立有待进一步研究。

Objective To explore the key technical problem of building the model of menopause using ovariectomized rats and the verifying of the model,and to provide valuable experience for the establishment of menopausal model.Methods Twenty 4-month female SD rats allocated to ovariectomy group（n=10）and mormal control group（n=10）.The rats in ovariectomy group were ovariectomized by longitudinal incision in the back.Rat exfoliated vaginal cells were examined by Giemsa staining,verify the model,and record the image.Levels of serum E2,FSH were determined by ELISA.The level of serum gonadal hormone were compared between two groups.Results Ten female rats were successfully ovariectomized,compared with the control group,the level of E2 in ovariectomy group decresed significantly,there were no significant changes of FSH.No incision infection occurred,one was found incision dehisced.Conclusion The method of building menopausal model has the advantage of small incision,simple operation and high success rate,further research work should be done in the sex hormone standard of ovariectomized rats.
