固相萃取－高效液相色谱法测定动物组织中甲氧苄氨嘧啶及磺胺类药物残留

Determination of Trimethoprim,sulfonamides in Animal Tissues by Solid Phase Extraction and High Performance Liquid Chromatography

本文建立了测定动物组织中甲氧苄氨嘧啶和五种磺胺含量的固相萃取-高效液相色谱方法。本方法以乙腈提取,稀盐酸反提,正己烷除脂肪,Oasis(R)HLB小柱净化,紫外检测器270 nm处进行检测。检测低限均能达到5μg/kg,线性在25μg/kg～300μg/kg良好,平均回收率77.50%～92.82%,RSD为3.34%～4.82%。该法具有准确度高、精密度好、检测限低等特点。

A solid phase extraction and high-performance liquid chromatography （HPLC） method was developed for the analysis of trimethoprim and 5 sulfonamides in animal tissues. Trimethoprim and sulfonamides were first extracted into acetonitrile and then back-extracted into dilute hydrochloric acid ,followed by liquid-liquid partition with n-hexane and cleaned-up on a Oasis（R） HLB cartridge. Samples were determined by HPLC with the variable wavelength detector （VWD）at 270 nm. Linearity of the method was good in the range of 25μ g/kg-300μ g/kg. The average recoveries were 77.50 %-92.82 %, the relative standard deviations were 3.34 %-4.82 % and the detection limit was 5μ g/kg.
