摘要
采用三角瓶摇床振荡培养,模拟序列间歇式活性污泥法(SBR)处理经微电解预作用后的皮革废水,考察驯化过程中微生物群落结构的变化情况.提取驯化过程中4个不同阶段混合菌群总DNA,采用降落式聚合酶链式反应和变性梯度凝胶电泳技术,对细菌16S rDNA V3区进行扩增和产物分离,并比较驯化过程中的微生物群落结构.结果表明,微生物群落结构和种群数量在驯化过程中存在明显的演替过程,不同微生物种群通过协同和竞争作用,最终形成一种具有新功能,性质稳定的群落结构.
This article used flask shaking experiment to imitate treating micro-electrolys pre-acted tannery wastewater by sequencing batch reactor(SBR).The diversity of the culture of microbial community in acclimation process was investigated.The genomic DNA of microbial community in four different periods was extracted.The bacterial 16S rDNA V3 fragment was amplified by touch-down polymerase chain reaction(PCR) method and the products were separated by denaturing gradient gel electrophoresis(DGGE).The results showed that there was an obvious syllogistic process of community structure and amount of dominant populations.A stabilizatied community structure with new function was formed at last through a series of cooperation and competition effects among each kind of microbe.
