摘要
目的研究瘦素在小鼠心肌缺血再灌注(MIR)损伤过程中的作用。方法24只雄性C57BL/6小鼠随机分为4组(每组6只)。(1)假手术组,(2)心肌缺血再灌注组(MIR组),(3)瘦素预处理组(Leptin+MIR组):用小鼠重组瘦素(recombinant mouse leptin,50μg/kg)在MIR手术前30min作腹腔注射制作而成,(4)瘦素术后处理组(MIR+Leptin组):用相同剂量瘦素于MIR手术后30min腹腔注射制作而成。术后6h采集各实验组标本,分别观察MIR小鼠血清和心肌瘦素的水平,并测量和计算心肌梗死面积。结果瘦素预处理可缩小小鼠心肌梗死面积,显著抑制MIR小鼠血清和心肌瘦素水平的升高。结论瘦素参与了小鼠MIR心肌损伤过程。瘦素预处理可降低MIR小鼠术后瘦素水平,减轻缺血再灌注损伤。
Objective To investigate tile role of leptin in myocardial ischemia reperfusion injury (MIRI)using a mouse MIRI model. Methods Twenty - four male C57BL/6 mice were randomized into 4 groups : (a) sham - operation group, (b) myocardial ischemia reperfusion group ( MIR group), (c) leptin pre - treatment group( Leptin + MIR group) and (d) leptin post -treatment group (MIR + Leptin group). MIR injury model was established by ligation of left coro- nary artery anterior descending branch for 30 min followed by repeffusion. Fifty ? g/kg of leptin was injected intraperito- neally 30 min before operation in leptin - pretreatment group and 30 rain after reperfusion in leptin post - treatment group. Samples of the blood and heart were harvested at 6 hour for analysis of myoeardial function and infarct size. Results Pretreatment of leptin significantly reduced myocardial infarction size and leptin levels in serum and myocardial cells in MIR injury model. Conclusion Leptin pretreatment shows protective effects on mouse MIRI by ameliorating the up - reg- ulation of leptin in serum and myocardium.
