摘要
目的:利用数据库信息获取人体组织microRNAs(miRNAs)表达数据,通过对其系统分析获得有益信息,进而为深入研究特定miRNA的功能提供新思路。方法:从miRNAs表达谱数据库获取人体多种组织miRNAs表达信息,按表达丰度、表达非特异性和特异性等对miRNAs表达谱进行分析;选择表达丰度大于0.004的miRNAs作为分析对象,并将在多种组织中均表达者定义为非特异性表达,而在单一组织中表达者则定义为特异性表达。结果:发现人体各种组织中miRNAs的表达谱存在明显差异,提示miR-122和miR-7两种miRNAs可能分别在肝脏和垂体中具有重要生理功能。结论:通过对人体miRNAs表达谱数据库资源的综合分析,有助于全面了解和掌握miRNAs在人体各种组织中的表达信息,并能为关键性miRNAs分子的筛选提供指导。
Objective:To obtain useful information via systematic analysis of human tissue miRNAs expression data,so as to support further researches on specific miRNAs.Methods:Human tissue miRNAs expression data were retrieved from five online miRNAs databases.A comprehensive analysis of their expression abundance and tissne specificity was carried out.Then,250 miRNAs with expression abundance more than 0.004 were selectecl as the objects of study.Those expressed in varied tissues were defined as tissue non-specific;those expressed only in one tissue were defined as tissue specific.Results:miRNAs expression profiles of eight human tissues were obtained,and the importance of some miRNAs was suggested by these miRNAs expression profiles.miRNA-122 and miRNA-7,for instance,are likely to play an important role in liver and pituitary respectively.Conclusion:It is an effective contrivance to study miRNAs by conducting a comprehensive analysis of miRNAs expression profiles from database resources.
