人宫颈癌HeLa细胞核内M-CSF高表达对抗肿瘤药物的影响 被引量:2

Effects of Nuclear M-CSF on the Sensitivity of Anti-tumor Chemotherapeutic Drug in Human Cervical Carcinoma HeLa Cells

摘要 目的探讨人宫颈癌HeLa细胞核内M-CSF高表达对抗肿瘤药物的影响。方法体外培养转染M-CSF人宫颈癌HeLa-M细胞、HeLa细胞、转染M-CSF空载体人宫颈癌HeLa-C细胞;分别以顺铂(0.2、2.0、20μg/mL)、紫杉醇(0.2、2.0、20μg/mL)作用于HeLa-M细胞、HeLa细胞、HeLa-C细胞48 h后,MTT法检测细胞相对活性;Western blot分析胞核内M-CSF对人宫颈癌HeLa细胞Bcl-2蛋白表达的影响。结果MTT比色法测定结果显示:在顺铂和紫杉醇单药药物浓度为20、2.0μg/mL时,HeLa-M细胞相对活性明显高于HeLa细胞、HeLa-C细胞(P<0.01),而在两药单药药物浓度为0.2μg/mL时三种细胞相对活性相比无明显差别(P>0.05)。HeLa细胞与HeLa-C细胞相比无明显统计学差异(P>0.05);Western blot法检测胞核内M-CSF明显上调人宫癌HeLa细胞Bcl-2蛋白的表达(P<0.01)。结论胞核内M-CSF能增加人宫颈癌HeLa细胞的药抗性并抑制HeLa细胞凋亡。 Objective To investigate effects of nuclear M - CSF on the sensitivity of anti - tumor chemotherapeutic drug in human ceiwical carcinoma HeLa cells. Methods Nuclear in M - CSF - transfected HeLa cells ( referred as to HeLa- M cell), pCMV,cyto,myc transfected HeLa cells (referred as to HeLa- C cell) or untransfected HeLa cells (referred as to HeLa cell) were cultured in vitro. After Treating the HeLa - M cells,HeLa - C ceils and HeLa cells with cisplatin(0.2,2.0,20 μg/mL)or paelitaxel( 0.2,2.0,20 μg/mL)for 48 h, the cells viability was determined using MTT assay. Western Blot was used to detect Bcl - 2 expression. Results The MTT assay showed that cell viability inhibition in HeLa - M cells treated by cisplatin and paclitaxel (20 μg/mL or 2.0 μg/mL) b-ere more than HeLa - C cell and HeLa cell ( P 〈 0.01 ). However, at 0.2 μg/mL there were no defference in the three kind of cells ( P 〉 0.05 ). Nuclear M - CSF up - regulated the expression of Bcl - 2 expression in HeLa cell( P 〈 0.01 ). Conclusion Nuclear M - CSF enhances chemotherapeutic drug resistance and apoptotie resistance of HeLa cells.
