摘要 目的研究Survivin、Caspase-3、cyclinD1在骨肉瘤中的表达及其临床意义。方法应用免疫组化S-P法检测36例骨肉瘤中Survivin、Caspase-3、cyclinD1的表达水平以及Survivin与Caspase-3、cyclinD1两者之间的相关性,并对所有病例进行随访。10例骨软骨瘤为对照组。结果①Survivin在骨肉瘤中的阳性率为63.7%(23/36),明显高于对照组骨软骨瘤10.0%(1/10)的阳性率,两者相比差异有统计学意义(P<0.01);Caspase-3在骨肉瘤中的阳性表达率为52.8%(19/36),而在骨软骨瘤中为90.0%(9/10),两者相比差异有统计学意义(P<0.01);cy-clinD1蛋白在骨肉瘤中的阳性率为74.4%(29/36),高于在骨软骨瘤中的40.0%(4/10),两者相比差异有统计学意义(P<0.05);前2者表达部位在细胞浆,cyclinD1表达于细胞核。②Survivin和Caspase-3表达强度之间呈显著负相关(r=-0.46,P<0.01)。③Survivin与cyclinD1表达呈正相关(r=0.641,P<0.01)。结论①Survivin在骨肉瘤组织中过表达与骨肉瘤的发生有关。②Caspase-3在骨肉瘤组织中低表达,Survivin可能通过抑制Caspase-3活性从而发挥其抑制骨肉瘤细胞的凋亡。③Survivin和cyclinD1在骨肉瘤的发生发展中可能起协同作用。 Objective To investigate the expression of Survivin,Caspase-3 and cyclinD1 in osteosarcoma and its clinical signficance. Methods Using immunohistoehemistry (S-P method) to detect the expressions of Survivin,Caspase-3 and cyclinD1 in 36 osteosarcoma tissues and 10 ostanchondroma tissues. Results (1)The expression rate of Survivin was significantly higher in osteosarcoma (63.7% ,23/36) than that in osteochondroma(10.0%, 1/10)(P〈0.01). The expression of Caspase-3 was detected in a greater proportion of osteochondroma(90.0% ,9/10) than that in osteosareoma(52.8%, 19/36)(P〈0.01). The positive expression rate of cyclinD1 was higher in osteosareoma(74.4% ,29/36) than that in osteochondroma(40.0% ,4/10). The former two are expressed in the cytoplasm,cyclinD1 is expressed in the nucleus. (2)There was a negative correlation was observed between the Survivin and Caspase-3 expressions in osteosarcoma(r=-0.46,P〈0.01). (3) There was a significantly positive correlation between Survivin and cyclinD 1 (r = 0. 641, P〈 0. 01). Conclusion (1) Survivin is over expressed in osteosmcoma. (2) Survivin exert probably the act of restraining oseteisarcoma cell apoptosis by the pathway of Caspase-3. (3)Survivin and cyclinD1 light play synergetie roles in the process of osteosarcoma.
