摘要
目的观察芪苈强心胶囊对H_2O_2诱导的H9C2大鼠心肌细胞凋亡的影响,探讨芪苈强心胶囊抗心力衰竭的作用机制。方法H9C2大鼠心肌细胞分为正常对照组、模型组、芪苈强心组,后2组采用100μmol/L H_2O_2处理6 h造成心肌细胞损伤模型,芪苈强心组加入含药血清,3组继续培养12、24、48 h,MTT法测定细胞增殖率。各组给药孵育48 h后,收集细胞,FCM方法测定细胞凋亡率,RT-PCR和Western blot法检测Caspase-3、Bcl-2、Bax、Fas/FasL mRNA和蛋白的表达。结果与模型组比较,芪苈强心组能增加H9C2细胞增殖活性,RT-RCR和Western blot方法结果显示芪苈强心组细胞凋亡率、Caspase-3、Bax、Fas、FasL表达明显下降,Bcl-2、Bcl-2/Bax明显上升。结论芪苈强心胶囊能下调Caspase-3、Fas/FasL表达,调节Bcl-2/Bax平衡,抑制心肌细胞凋亡,对心肌细胞起到一定的保护作用。
Objective To investigate the effects of traditional Chinese medicine Qiliqiangxin capsule on H9C2 myocardial cell apoptosis induced by H2O2 in rats,and further to elucidate the molecular mechanisms.Methods The H9C2 myocardial cells were divided into the normal group,model group,Qiliqiangxin capsule treatment group. The model was established by injuring myocardial cell with 100μmol/L H2O2 for 6h.Each group were cultivated and measured the cell proliferation ratio by MTT method at different time points(12,24 and 48h).Cells were collected after 48h and subjected to RT-PCR and Western blot assays for the expression levels of Caspase-3,Bcl-2,Bax,Fas/FasL mRNA and protein.The percentage of apoptosis cells were detected by FCM.Results Compared with the model gronp,the rates of cell growth were significantly increased while the percentage of apoptosis cells were deceased in Qiliqiangxin capsule treatment group.The results of RT PCR and Western blot demonstrated that the expression of Caspase-3,Bax,Fas/FasL were decreased,and the expression of Bcl-2,Bcl-2/Bax were increased.Conclusion The traditional Chinese medicine Qiliqiangxin capsule could protect H9C2 from apoptosis through decreasing the expression of Caspase-3,Fas/FasL,and regulating the balance between Bcl-2 and Bax.
