加味桃核承气汤对肝纤维化大鼠TGF-β1蛋白表达的影响

To Study the Effects of TGF-β1 of Jiaweitaohechengtang on Hepatic Fibrosis Induced by CCl_4 in Rats

[目的]观察加味桃核承气汤对四氯化碳肝纤维化大鼠肝组织转化生长因子-β1(TGF-β1)蛋白表达的影响。[方法]用四氯化碳诱导大鼠肝纤维化模型,研究分组为正常对照组、模型对照组、加味桃核承气汤高、中、低剂量治疗组和秋水仙碱对照组;灌胃给药,疗程8周;用免疫组化技术检测肝组织TGF-β1的蛋白表达;检测血清ALT、AST水平;用苦味酸-天狼红胶原染色方法进行半定量肝纤维化计分(SSS)。[结果]高、中、低剂量组及秋水仙碱组的TGF-β1的表达水平均低于模型对照组;经加味桃核承气汤治疗后血清ALT、AST水平及肝纤维化SSS计分显著下降。[结论]加味桃核承气汤可抑制肝纤维化大鼠TGF-β1的表达,达到抗肝纤维化作用。

[bjective]To study the effects of TGF β1 of Jiaweitaohechengtang on hepatic fibrosis induced by CCl4 in rats. [Methods] Hepatic fibrosis models were induced by CCl4 in rats. All the experimental rats were divided into six groups： normal control group, model control group, colchicine positive treatment group, high-dose, medium dose, lowdose jiaweitaohechenqitang treatment groups. Drugs were fed on rats for 8 weeks by intragastric administration. Protein expression levels of transfer growth faetors-β1 （TGF-β1）were determined with immunohistochemical staining method. Scores of hepatic fibrosis were assessed with semiquantitive scoring system（SSS）through tianlanghong staining. [Results]The protein expression levels of TGF-β1 in the liver tissues in model control group were markedly increased compared with those in normal control group. Scores of hepatic fibrosis and the levels of serum ALT and AST in model control group were higher than those in normal control group, but lower than those in three treatment groups with jiaweitaohechengqitang. There were significant differences between model control group and three jiaweitaohechengqitang treatment groups. [Conclusion]Jiaweitaohechengqitang has therapeutic effects on hepatic fibrosis induced by CCl4 in rats.
