逐痰通络汤对急性期脑出血大鼠血IL-1β、IL-6表达的影响 被引量:2

Effect of Zhutan Tongluo Tang on the Expression of IL-1β and IL-6 in Acute Cerebral Hemorrhage Rat

摘要 [目的]探讨中药逐痰通络汤对脑出血损伤的保护作用。[方法]将大鼠随机分为正常对照组(N组)、假手术组(K组)、模型组(C组)和给药组(Z组),采用胶原酶诱导法建立大鼠脑出血模型,观察各组动物神经功能缺损并评分,ELISA法检测血清IL-1β、IL-6的表达。[结果]①通过神经功能评分,Z组与C组比较,第1、2d无明显统计学差异,第3d有差异(P<0.05),评分较低,第7d有显著差异(P<0.01);②与K组比较,C组IL-1β、IL-6含量在第1d时开始升高(P<0.05),第2、3d到达高值(P<0.001),第7d接近正常范围;③给药后,第2d,Z组IL-1β、IL-6含量较C组明显下降(P<0.05),第3d,显著下降(P<0.001)。[结论]逐痰通络汤能抑制IL-1β、IL-6的表达和释放,从而减轻炎症反应,对脑出血后的继发性神经元损伤起保护作用。 [Objective] To investigate the effect of Zhutan Tongluo Tang on the expression of interleukin-1β(IL-1β) and interleukin-6(IL-6) and changes of nerve function in acute cerebral hemorrhage rat.[Methods] Rats were randomly divided into four groups: normal control group(N),sham-operative group(K),model group(C) and Zhutan Tongluo Tang group(Z),Collagenase was used to establish rat model of intracerebral hemorrhage,each group of animal nerve function damage was evaluated and graded after operation,ELISA method was used to test the expression of IL-1β and IL-6 in blood.[Results] 1.Score of nerve function: there was no significant difference between Z and C group in the first 1,2d.There was difference on 3d(P0.05),and significant difference on 7d(P0.01).For Z group had lower score with C group;2.The level of IL-1β and IL-6 began to increase in C group on 1d,compared with K group(P 0.05),and reach high level in the first 2,3d(P 0.001),the level came to normal range on 7d;3.After administration,compared with C group,the level of IL-1β and IL-6 began to decrease on 2d(P 0.05),and decreased significantly on 3d(P 0.001).[Conclusion] Zhutan Tongluo Tang can depress the expression and release of IL-1β and IL-6,inhibiting inflammation response,which has an evident protective effect on secondary injured neuron after intracerebral hemorrhage in rats.
