柴郁温胆汤及其拆方对抑郁大鼠血清ACTH、COR及海马cAMP的影响 被引量:21

摘要 [目的]探讨柴郁温胆汤及其拆方对抑郁大鼠血清ACTH、COR和海马cAMP的影响。[方法]健康雄性SD大鼠84只,随机分为正常组、模型组、柴郁温胆汤组、化痰组、调气血组、养心脾组、马普替林组。参照Katz刺激方法制备慢性轻度不可预见性应激(CUMS)抑郁模型。采用放射免疫法测血清ACTH、COR和大鼠海马cAMP含量。[结果]与正常组相比,模型组大鼠血清ACTH和COR水平明显升高、海马cAMP含量明显降低。同模型组相比,柴郁温胆汤组及调气血组能明显降低ACTH浓度(P<0.05);柴郁温胆汤组、化痰组和调气血组均能显著降低COR浓度(P<0.01);柴郁温胆汤组、化痰组和马普替林组均能够显著提升大鼠海马cAMP的含量(P<0.01)。[结论]慢性轻度不可预见性应激可以导致大鼠血清ACTH和COR水平升高;海马cAMP含量下降,柴郁温胆汤可以逆转这种改变,其中以调气血和化痰药作用为主,其机制可能与其参与调节HPA轴功能和细胞内信号传导有关。 [Objective] To explore the effects of the traditional Chinese drugs Chaiyuwendantang(CYWDT) and its component on the plasm levels of ACTH,COR and cAMP in hippocampus of depressed mode.[Methods] Eighty——four male rats were divided into seven groups: control group,depressed model group,group of CYWDT,group of HuaTan,group of TiaoQiXue,group of YangXinPi,group of maprotiline equally and made up models in chronic unpredictable mild stress(CUMS).The plasm levels of ACTH,COR and the level of cAMP in hippocampus were detected by radioimmunoassay.[Results] We found that the plasm levels of ACTH,COR were significantly increase and cAMP in hippocampus was significantly decreasd in depression model of rat.CYWDT and TiaoQiXue could significantly decreased the level of ACTH(P0.05);CYWDT,HuaTan and TiaoQiXue could significantly decreased the level of COR(P0.01).CYWDT,HuaTan and TiaoQiXue could significantly increase the level of cAMP in hippocampus.[Conclusion] Chronic unpredictable mild stress can increase the plasm levels of ACTH,COR and the level of cAMP in hippocampus of depression rats.CYWDT could reverse all the changes.The mechanism maybe relatd to regulate hypothalamic-pituitary-adrenal axis and signal transmission in cell.
