The influence of age on the levels of Rab3a in different brain regions in SAMP8 mice

摘要 目的探讨年龄对小鼠不同脑区Rab3a含量的影响。方法77只SAMP8小鼠分为4个年龄组:5、9、13、15月龄组。在进行行为学特征化后采用免疫印迹技术检测Rab3a蛋白和内参蛋白GAPDH在背海马、腹海马、额叶、顶叶和嗅球中的表达,Rab3a灰度值与GAPDH灰度值的比值作为Rab3a的相对含量。结果在腹海马中,15月龄SAMP8小鼠Rab3a的相对含量显著低于5、9、13月龄鼠(P值分别为0.014,<0.001及0.001),而背海马、额叶、顶叶和嗅球Rab3a含量在各个年龄段间差异均无统计学意义(P值均>0.05)。结论在生命过程中,SAMP8小鼠脑内Rab3a含量相对恒定,仅高龄小鼠有区域特异性降低。 Objective To explore the effects of age on the levels of Rab3a in different brain regions in SAMP8 mice.Methods 77 SAMP8 mice were consisted of four age groups:5,9,13 and 15 months respectively.After detected the ability of learning and memory by Morris water maze,the Rab3a and GAPDH (reference protein) in dorsal hippocampus,ventral hippocampus,frontal lobe,parietal lobe and olfactory bulb were detected by Western blot,and the ratio of the Rab3a to the GAPDH gray-scale value was viewed as the relative content of the Rab3a.Results The relative amount of the Rab3a in the 15-month-old group was significantly lower than those in the 5-,9-and 13-month old mice in the ventral hippocampus (the values of P were 〈0.014,0.001 and 0.001,respectively).However,the amounts of Rab3a were similar in the dorsal hippocampus,frontal lobe,parietal lobe and olfactory bulb among the groups (P〈0.05).Conclusion During the lifetime,the amount of Rab3a is relatively preserved in the brain of SAMP8 mice,and decreases only in the elder mice in a region-specifical manner.
