多重PCR法对乳制品中3种致病菌的同时快速检测 被引量:6

摘要 通过调整混合培养基的配比摸索乳制品中金黄色葡萄球菌、沙门氏菌和志贺氏菌的快速共增菌条件,并选择稳定高效的细菌基因组DNA提取方法,然后根据三种目标致病菌的保守基因分别设计多重PCR引物,对致病菌进行多重PCR检测。结果表明,方法具有良好的针对目标致病菌的特异性,检测限可达102 mL-1,且反应体系各组分间未见交叉干扰。对人工污染的乳品盲样检测验证了方法的实用性。 In the present study,we first optimized conditions for simultaneous enrichment of Staphylococcus aureus,Salmonella spp.and Shigella spp.in dairies,and then selected a stable and efficient method to extract genomic DNA.Subsequently,a multiplex PCR approach was settled,aiming at the conserved genes of target pathogens,respectively.This multiplex RTi-PCR procedure was verified to be specific to the target bacteria without cross interference,and the detection limit was 10^2 mL^-1 for each pathogen.Finally,it was tested in artificially inoculated dairies,revealing a rapid,sensitive and accurate detection of the target bacteria.
