摘要
目的检测寻常型银屑病皮损中树突细胞溶酶体相关膜蛋白(DC-LAMP)、树突细胞特异性非整合素(DC-SIGN)的表达,探讨其在银屑病发病机制中的作用。方法采用免疫组化SP法检测DC-LAMP、DC-SIGN在寻常型银屑病皮损与正常皮肤的表达。结果DC-LAMP在银屑病表皮角质形成细胞(KC)基底层和棘层以及真皮树突细胞(DC)均高表达,在正常皮肤表皮KC基底层和真皮DC中呈低表达(P<0.01);DC-SIGN在银屑病表皮KC和真皮DC中均高表达,在正常皮肤表皮KC基底层和真皮DC中低表达(P<0.01)。结论寻常型银屑病发病与DC-LAMP、DC-SIGN有关。DC、KC可能通过表达DC-LAMP和DC-SIGN发挥抗原递呈作用,参与银屑病免疫发病过程。
Objective To investigate expression of DC-lysosomal-associated membrane protein(DC-LAMP)and Dendritic-cell specific ICAM-3 grabbing non-integrin(DC-SIGN) on Dendritic cells in Psoriasis vulgaris,and to explore the role of DC-LAMP and DC-SIGN in Psoriasis vulgaris. Methods Expressions of DC-LAMP and DC-SIGN in lesions of Psoriasis vulgaris and normal skin were detected by immunohistochemistry. Results DC-LAMP were highly expressed in keratinocytes in the basal layer and spinous layer of the epidermis and Dendritic cells in the dermis of Psoriasis vulgaris,while lowly expressed in the basal layer of the epidermis and Dendritic cells in the dermis of normal skin (P〈 0. 01). DC-SIGN were highly expressed in keratinocytes of Psoriasis vulgaris and Dendritic cells in the dermis, while lowly expressed in keratinocytes in the basal layer of the epidermis and Dendritic cells in the dermis of normal skin (P〈 0. 01). Conclusion The pathogenesis of Psoriasis vulgaris may be related with DC-LAMP and DC-SIGN. Denritic cells and keratinocytes might play antigen-presenting roles by expressing DC-LAMP and DC-SIGN involved in the immune reaction of Psoriasis.
