尖锐湿疣组织中TGFβI、TGFβRⅡ及cyclin E的表达

摘要 目的:检测转化生长因子βⅠ(TGFβⅠ)、转化生长因子βⅡ受体(TGFβRⅡ)和细胞周期素E(cyclin E)蛋白在尖锐湿疣(CA)组织中的表达。方法:采用免疫组化SP法和图像分析系统检测30例CA患者皮损,15名正常人皮肤(包皮)组织中TGFβⅠ、TGFβRⅡ及cyclin E的表达。结果:TGFβI和cyclin E在CA组织中的表达均显著高于正常皮肤组织(P<0.05);TGFβRⅡ在CA组织中的表达显著低于正常皮肤组织(P<0.05);TGFβⅠ与cyclin E在CA组织中的表达呈正相关(P<0.05);TGFβRⅡ与cyclin E在CA组织中的表达呈负相关(P<0.05)。结论:TGFβRⅡ表达下调,使TGFβ信号传导通路受阻,降低了TGFβⅠ对cyclin E表达的抑制作用,及其对细胞增殖的负性调节作用,使CA组织过度增殖,可能是CA发病机制之一。 Objective: To investigate the expression of transforming growth factor βI (TGFβI), transforming growth factor- β II Receptor (TGFβRII)and cyclin E in condyloma acuminatum (CA). Methods: Immunohistochemical SP method and color pathological image computer analysis system were applied to detect the expressions of TGFβI, TGFβRII and cyclin E in 30 patients with CA and 15 normal human skin samples (foreskin). Results: The expression of TGFβI and cyclin E was significantly higher in CA than in normal foreskin tissue ( P 〈 0.05). The expression of TGFβRII was significantly lower in CA than in normal foreskin tissue (P 〈 0.05). The expression of TGF131 was positively correlated to cyclin E ( P 〈 0.05) ; The expression of TGFβRII was negatively correlated to cychn E ( P 〈 0.05). Conclusion: The down - regulation of TGFβRII may disrupt the TGFβ signal transduction and disable the TGFβI negative regulation of cell proliferation, and then induce the over expression of cyclin E, which can disorder cell cycle control and promote the cell proliferation.
