骨涎蛋白单克隆抗体抑制亲骨转移人乳腺癌细胞血管侵袭机制的探讨 被引量:1

Inhibitory effects of bone sialoprotein monoclonal antibody on bone-seeking breast cancer cells intravasation into blood vessels

摘要 目的:探讨骨涎蛋白(BSP)单克隆抗体对亲骨转移人乳腺癌细胞株MDA-MB-231BO血管侵袭能力的影响。方法:MTT法检测BSP抗体对MDA-MB-231BO细胞的增殖作用;鸡胚尿囊绒膜(CAM)实验检测BSP抗体对MDA-MB-231BO细胞血管侵袭能力的影响。结果:BSP抗体对MDA-MB-231BO细胞的生长具有特异抑制作用,其抑制率和剂量呈正相关,BSP抗体浓度为100μg/mL时,抑制率为35.18%。BSP抗体能特异性抑制MDA-MB-231BO细胞穿透鸡胚血管内皮细胞和基底膜,抑制效应与抗体浓度呈正相关。100μg/mL的BSP抗体可以明显抑制MDA-MB-231BO细胞侵入鸡胚血管。结论:BSP抗体对MDA-MB-231BO细胞的血管侵袭能力具有特异抑制作用,提示BSP抗体可抑制乳腺癌靶向骨转移的进程。 OBJECTIVE:To determine the inhibitory effects of anti-bone sialoprotein (BSP) antibody on human breast cancer cells intravasation into blood vessels in vivo. METHODS:MTT assay was carried out to investigate the effect of anti-BSP antibody on proliferation of MDA-MB-231BO cells; Chick embyro chorioallantoic membrane (CAM) assay was carried out for determining the effect of anti-BSP antibody on MDA-MB-231BO cells intravasation into blood vessels. RESULTS:Anti-BSP antibody inhibited the proliferation of MDA-MB-231BO cells in a dose and time-dependent manner. When the concentration of anti-BSP antibody was up to 100 μg/mL,the inhibition rate was 35.18%. The anti-BSP antibody had specific inhibition effect on MDA-MB-231BO cells passing through chick embyro endothelial cells and basement membrane in a dose dependent manner. When the concentration of anti-BSP antibody was up to 100 μg/mL,it can obviously inhibited MDA-MB-231BO cells intravasation into chick embyro blood vessels. CONCLUSIONS:Anti-BSP antibody can specifically inhibit breast cancer cells intravasation into chick embyro blood vessels,which suggests that anti-BSP antibody may inhibit the metastasis cascade of breast cancer to bone.
