摘要
目的:探讨骨涎蛋白(BSP)单克隆抗体对亲骨转移人乳腺癌细胞株MDA-MB-231BO血管侵袭能力的影响。方法:MTT法检测BSP抗体对MDA-MB-231BO细胞的增殖作用;鸡胚尿囊绒膜(CAM)实验检测BSP抗体对MDA-MB-231BO细胞血管侵袭能力的影响。结果:BSP抗体对MDA-MB-231BO细胞的生长具有特异抑制作用,其抑制率和剂量呈正相关,BSP抗体浓度为100μg/mL时,抑制率为35.18%。BSP抗体能特异性抑制MDA-MB-231BO细胞穿透鸡胚血管内皮细胞和基底膜,抑制效应与抗体浓度呈正相关。100μg/mL的BSP抗体可以明显抑制MDA-MB-231BO细胞侵入鸡胚血管。结论:BSP抗体对MDA-MB-231BO细胞的血管侵袭能力具有特异抑制作用,提示BSP抗体可抑制乳腺癌靶向骨转移的进程。
OBJECTIVE:To determine the inhibitory effects of anti-bone sialoprotein (BSP) antibody on human breast cancer cells intravasation into blood vessels in vivo. METHODS:MTT assay was carried out to investigate the effect of anti-BSP antibody on proliferation of MDA-MB-231BO cells; Chick embyro chorioallantoic membrane (CAM) assay was carried out for determining the effect of anti-BSP antibody on MDA-MB-231BO cells intravasation into blood vessels. RESULTS:Anti-BSP antibody inhibited the proliferation of MDA-MB-231BO cells in a dose and time-dependent manner. When the concentration of anti-BSP antibody was up to 100 μg/mL,the inhibition rate was 35.18%. The anti-BSP antibody had specific inhibition effect on MDA-MB-231BO cells passing through chick embyro endothelial cells and basement membrane in a dose dependent manner. When the concentration of anti-BSP antibody was up to 100 μg/mL,it can obviously inhibited MDA-MB-231BO cells intravasation into chick embyro blood vessels. CONCLUSIONS:Anti-BSP antibody can specifically inhibit breast cancer cells intravasation into chick embyro blood vessels,which suggests that anti-BSP antibody may inhibit the metastasis cascade of breast cancer to bone.
