纳米羟基磷灰石-壳聚糖复合材料对NIH3T3细胞粘附增殖的影响 被引量:3

摘要 目的制备纳米羟基磷灰石-壳聚糖(nano hydroxyapatite-chitosan,nHA/CS)复合膜和三维立体支架,观察复合材料的两种状态对成纤维细胞粘附形态的影响。方法制备nHA/CS复合膜和HA/CS复合膜,然后将两组膜分别与NIH3T3细胞共培养,倒置显微镜下观察细胞在材料表面粘附形态同时进行粘附细胞数量的测定;冷冻干燥法制备nHA/CS三维支架,将HA制成同样大小的圆盘状,然后将NIH3T3细胞分别与两组材料共培养,扫描电镜观察细胞在两种材料表面的粘附形态同时进行细胞粘附数量的测定和能谱分析。结果与NIH3T3细胞共培养5、7d后nHA/CS组细胞粘附数量高于HA/CS组和HA组,差异有显著性。结论nHA/CS复合材料有促进细胞粘附增殖作用。 Objective To prepare nanometer hydroxyaptite/chitosan(nHA/CS)compound membrane and composite scaffold,and observe the adhesive quantity and behavior of NIH3T3 cells in the two states of compound.Methods nHA/CS compound membrane and HA/CS compound membrane were prepared and then co-cultured with NIH3T3 cells.The adhesive quantity and behavior of NIH3T3 cells in the two compound membranes were observed in upside-down microscope.The composite scaffold was prepared by lyopyilization,and HA was prepared in the same size and shape of disk.Co-cultured NIH3T3 with HA and nHA/CS,cells adhesive quantity was determined and morphology was examined with SEM and spectrum analysis.Results After 5 and 7 days,the adhesive quantity of cells on nHA/CS was higher than on HA/CS and HA.Conclusions The nHA/CS showed significant abilities of adherence and proliferation.
