摘要
目的:通过在缺氧条件下体外培养成骨细胞,探讨缺氧对成骨细胞增殖与凋亡的影响以及淫羊藿甙的保护机制。方法:采用酶消化法传代培养乳SD大鼠颅盖骨细胞并建立缺氧模型。实验分2组:组1为单纯缺氧条件下进行的细胞培养,不加入任何药物干预;组2为缺氧条件下加入浓度为10ng/ml的淫羊藿甙与细胞共培养,用台盼蓝染色法分别计数培养5d的细胞数;用流式细胞仪检测2组缺氧5d的细胞凋亡率。结果:缺氧组的细胞数明显少于与淫羊藿甙共培养组,缺氧引起的细胞凋亡率较淫羊藿甙共培养组高。结论:缺氧能抑制成骨细胞的增殖,促进成骨细胞调亡而淫羊藿甙对其具有保护作用。
Objective:To investigate the effects of hypoxia on osteoblasts proliferation and apoptosis and the protective effect of Icariin.Methods:The osteoblasts obtained from the calvaria of neonatal SD rats were cultured by enzyme digestion in a hypoxic environment,group 1 with no treatment and group 2 co-cultured with 10ng/ml Icariin.Trypan blue staining was used to measure the cell number and flow cytometer for the rate of apoptosis in both groups on the 5th day after culture.Results:The cell number of hypoxic group was fewer than that of contrast group,and the rate of apoptosis in hypoxic environment was higher than contrast group.Conclusion:Hypoxia can inhibit the proliferation of osteoblasts and promote the apoptosis of osteoblasts.Icariin can protect osteoblasts from apoptosis.
