昆明种小鼠单次肌肉注射溶瘤腺病毒注射液SG600-p53的急性毒性试验

摘要 目的观察肿瘤特异性溶瘤腺病毒SG600-p53注射液给予昆明种小鼠最大体积和浓度的条件下不产生死亡的最大耐受剂量(MTD)。方法SG600-p53注射液单次肌肉注射给予小鼠,剂量为2.5×1013vp/kg,给药容量为25ml/kg,给药后连续观察2周。结果给药后小鼠主要中毒表现为精神萎靡,活动减少,双眼微闭,注射后约10~30min后恢复。试验期间,给药组的动物体重增长缓慢,食量减少。在整个试验过程中给药组和对照组动物均未出现死亡。结论SG600-p53注射液对小鼠有一定的轻微毒性,其MTD为2.5×1013vp/kg。 Objective To evaluate the maximal tolerated dose(MTD)of the oncolytic adenovirus SG600-p53 which was injected into Kunming mice with the maximal volume and concentration and did not result in mouse death.Methods Mice were injected SG600-p53 by single intramuscular injection with the dose of 2.5×1013vp/kg and volume of 25ml/kg,and then were observed for 2 weeks.Results After injection,mice showed some toxic behaviors,e.g.psychic listlessness,decreased activity and eye closure.These mice recovered after 10-30min.The body weight of mice in the experimental group was increased slowly,mouse food consumption was decreased obviously,and no mice died during the observation period.Conclusion The oncolytic adenovirus SG600-p53 has some slight toxicity in mice and its MTD is 2.5×1013vp/kg.
