摘要
为了获得纺织工业中所需求的高温退浆酶,从枯草堆、农田积肥、下水道等处土样中分离得到1株能向胞外分泌耐高温α-淀粉酶的菌株并对其产酶酶学性质进行研究。研究结果表明:该菌为枯草芽孢杆菌(Bacillus subtilis),初步命名为Bacillus subtilisC1;菌株C1在基础培养基中(pH=7,温度45℃)培养24h左右达到生长稳定期,在68h时,酶活达到最高,为185.6U/mL;此α-淀粉酶最适作用pH值为6,最适温度为70℃;酶的耐热性能较好,在70℃处理1h,仍具有81%的酶活,适合高温退浆的需要。
In order to obtain the therrnostable desizing enzyme, which can be used in textile industry, a strain which can excrete thermostable a-amylase was isolated from soil, and the characteristics of its crude enzyme were studied. The results show that the strain is identified preliminarily as Bacillus subtilis, temporarily named Bacillus subtilis C1. In the basal medium, the strain C1 reaches its stationary phase within 24 h at pH 7 and 45 ℃. And at 68 h, it reaches the highest enzyme activity of 185.6 U/mL. The optimum pH value is 6 and the optimum temperature is 70℃. This enzyme is stable to high temperature, and 81% of enzyme activity is retained after being disposed at 70 ℃ for 1 h.
