摘要
目的:研究脑缺血后STAT1mRNA在病灶局部的表达水平以及电针的干预影响。方法:通过电凝闭大鼠大脑中动脉致局灶性脑缺血模型,采用原位杂交的方法检测不同时间段STAT1mRNA在病灶侧皮质、海马CA1区的表达水平以及电针的干预效果。结果:模型2h组无显著性变化;模型24h组多个指标稍有增高;模型组3d组指标均有显著的增加。电针组中电针2h组、电针24h与模型24h组比较,差异无显著性意义(P>0.05);电针3d组与模型3d组比较,所有指标均明显偏低,差异均有非常显著性意义(P<0.01)。结论:脑缺血3天后STAT1mRNA表达水平明显增加,而电针治疗对其有一定的抑制作用。
Objective:To study STAT1 mRNA expression in rat models of focal cerebral ischemia and to observe the influence of electroacupuncture intervention.Methods:Heat coagulation of rat middle cerebral arteries was used for the establishment of focal cerebral ischemia model.In-situ hybridization method was used for the detection of STAT1 mRNA expression in the affected cortex and hippocampal CA1 region.Results:No obvious change was found in 2h model group.Most of the observation indexes were increased in 24h model group,and all of the indexes were markedly increased in 3d model group.The difference between 2h electroacupuncture group,24h electroacupuncture group and 24h model group was insignificant (P > 0.05).The indexes were lower in 3d electroacupuncture group than those in 3d model group(P < 0.01).Conclusion:Three days after cerebral ischemia,STAT1 mRNA expression is presented in rat models of focal cerebral ischemia,and electroacupuncture has an inhibition on STAT1 mRNA expression.
