摘要
目的观察中药牛黄天龙方药对荷瘤小鼠的抗肿瘤作用及造血系统的影响。方法取BALB/C小鼠100只,随机分为牛黄天龙方药高、中、低剂量组,模型组及环磷酰胺组,采用皮下注射宫颈癌细胞株的方法建立U14荷瘤小鼠模型,分别给予牛黄天龙方药8、6、4g/(kg.d)及0.9%氯化钠注射液灌胃、环磷酰胺腹腔注射。用药10d后,取50只小鼠取血处死后计算抑瘤率和外周血白细胞及骨髓细胞计数;另50只继续饲养观察存活时间。结果各治疗组瘤质量、生存时间与模型组比较差异均有统计学意义(P<0.05),瘤质量均低于模型组而生存时间均长于模型组,牛黄天龙方药大、中、小剂量组生存时间均长于环磷酰胺组(P<0.05);牛黄天龙方药大、中、小剂量组外周血白细胞计数高于环磷酰胺组、低于模型组(P<0.05),牛黄天龙方药大、中、小剂量组骨髓细胞计数高于环磷酰胺组(P<0.05),与模型组比较差异无统计学意义(P>0.05)。结论牛黄天龙方药具有显著的抗肿瘤作用且对造血系统的影响较小。
Objective To observe Niuhuang Tianlong prescription on the antitumor effect and hematopoietic system in tumor - bearing mice. Methods U14 tumor - bearing model mice were established by using BALB/C mice and hypodermic of uterine cervix cancer (UCC) cell strain. 100 model mice were randomly divided into high - . middle- , and low - dose Niuhuang Tianlong prescription groups, model group and Cyclophosphamide group. 8,6, 24 g/(kg · d), of Niuhuang Tianlong prescription,0.9% sodium chloride injection and Cyclophosphamide injection. Ten days after treatment, 50 mice were killed for calculating tumor inhibitory rate and counter of peripheral blood leucocyte and bone marrow cell. Survival time was observed in other 50 mice. Results The tumor inhibitory rate in each treatment groups were higher than that in model group ( P 〈 0.05 ). Survival time in each treatment groups were longer than that in model group ( P 〈 0.05 ). Survival time in high - , middle - , and low - dose Niuhuang Tianlong prescription groups were longer than that in Cyclophosphamide group ( P 〈 0.05 ). The counter of peripheral blood leucocyte in high - , middle - , and low - dose Niuhuang'Tianlong prescription groups were more than that in Cyclophosphamide group ( P 〈 0.05 ) , and less than that in model group ( P 〈 0.05 ). The counter of bone marrow cell in high -, middle -, and low - dose Niuhuang Tianlong prescription groups were more than that in Cyclophosphamide group ( P 〈 0.05). Conclusion Niuhuang Tianlong prescription has obvious antitumor effect and smaller effect on hematopoietic system.
