摘要
采用直接测序法检测LHβ基因在黔北麻羊中的单核苷酸多态性,以探讨促黄体素β亚基基因多态性与黔北麻羊产羔数的关系,以及该基因对黔北麻羊繁殖力的影响。结果表明:LHβ基因检测到2个突变位点,在外显子2第414位点发生了由C→T碱基突变,导致苏氨酸突变为甲硫氨酸;检测到AA、Aa 2种基因型。A等位基因频率为0.933 4,a等位基因频率为0.066 7,2种基因型之间产羔数差异均不显著(P>0.05)。在内含子2第718位点发生了由G→A碱基突变,检测到BB、Bb 2种基因型。B等位基因频率为0.950 0,b等位基因频率为0.0500,2种基因型之间产羔数差异均不显著(P>0.05)。
LHβ gene is regarded as candidate genes for the prolificacy. Single nucleotide polymorphism of LHβ gene was detected in Qianbei -pockmarked Goats by sequencing in the present work. The results revealed that there was two single nucleotide mutation (C^414→T^414) at exon 2 of LHβ gene in Qianbei - pockmarked Goats, which resulted in an amino acid change: Threonine-Methionine. Two genotypes (AA and Aa) was detected in Qianbei - pockmarked Goats. The frequency of allele A was 0.933 4, and that allele a was 0.066 7. No significant difference (P 〉0. 05) in litter size was obtained between two genotypes in Qianbei -pockmarked Goats. Another mutation of (G^718→A^718 ) at intron 2 of LHβ gene led to two genotypes (BB and Bb) and was also detected in Qianbei -pockmarked Goats. Frequency of allele B and b was 0.950 0 and 0.050 0, respectively. The litter size between two genotypes were also nonsignificant in Qianbei -pockmarked Goats (P 〉 0.05).
