摘要
应用单因子和正交两种优化方法,考察红曲霉Monascus菌株-ZZ产麦角固醇皂化的最佳条件。单因子实验结果表明:最佳皂化条件碱体积分数为20%,醇碱比为2∶1,物料与碱的比例为1∶1,皂化时间为2 h,皂化温度为90℃。正交优化结果显示:不同因子对麦角固醇提取的影响顺序是皂化温度>皂化时间>物料皂化剂比>碱体积分数>醇碱比;最佳条件是物料皂化剂比为1∶1,碱体积分数为15%,醇碱比为4∶1,皂化温度为90℃,皂化时间为3 h。
The single factor test and orthogonal experiments had been carried out to optimize the condition of ergosterol saponification from the Monascus sp. ZZ mycelium. The result of single factor test indicated that the optimal conditions were 20% of caustic soda concentration, 2:1 of ethanol to alkali, 1 : 1 of sample to the alkali, and saponification at 90℃ for 2 h. Orthogonal test showed that the effect order among the factors were the saponified temperature 〉 the saponified time 〉 the ratio of sample to saponifier 〉 the caustic soda concentration 〉 the ratio of ethanol to alkali. The optimal ratio of sample to saponifier, caustic soda concentraratio of ethanol to alkali, saponified temperature, and saponified time are 1: 1, 15%, 4: 1,90℃, 3 h,respectively.
