摘要
应用超声波直接转化法、农杆菌介导转化法、恒定频率的超声波仪处理植物15 min后再用农杆菌介导法侵染植物和在农杆菌侵染过程中辅助以超声波处理10 s 4种方法对鱼腥草进行转基因研究,结果显示:上述4种转化法对gus基因最大瞬时表达率依次为47%、60%、80%和75%。结论:用恒定频率的超声波仪处理植物15 min后再用农杆菌介导法侵染植物和在农杆菌侵染过程中辅助以超声波处理10 s两种转化法gus基因的瞬时表达率最高,说明超声波辅助农杆菌介导遗传转化方法是一种高效的鱼腥草转基因方法,它实现了外源基因在植物中的高效表达。
In this study,4 kinds of methods(sonication direct conversion method,Agrobacterium-mediated transformation method,Agrobacterium-mediated infection to plants after being treated with constant-frequency sonication instrument for 15 min,and sonication treatment for 10 s in the process of Agrobacterium infection to plants) were used to study the gene transformation of Houttuynia cordata Thunb,the results showed that the maximum transient expression rate of gus gene transformed by the above 4 kinds of methods was 47%,60%,80% and 75% respectively.Therefore sonication-assisted Agrobacterium-mediated genetic transformation method is an efficient method for the genetic transformation of Houttuynia cordata,it can realize the high-efficient expression of exogenous genes in plants.
