血管紧张素Ⅱ对体外培养Ins-1细胞作用的研究

Effects of Angiotensin Ⅱ on Ins-1 cells cultured in vitro

目的观察血管紧张素Ⅱ对体外培养的Ins-1细胞增殖及凋亡的作用。方法 :将血管紧张素Ⅱ和体外培养的Ins-1细胞共培养后,应用细胞增殖试验方法测定Ins-1细胞的增殖率,用流式细胞检测方法检测Ins-1细胞的早期凋亡率。结果血管紧张素Ⅱ在和体外培养的Ins-1细胞共培养12 h后,Ins-1细胞的增殖率明显下降,凋亡率明显增高,且与血管紧张素Ⅱ浓度相关。结论血管紧张素Ⅱ对体外培养的Ins-1细胞在特定浓度和时间的条件下有抑制细胞增殖,诱导凋亡增加的作用。

Objective To observe the apoptosis-induced and proliferation-restrained effects of angiotensin Ⅱ on Ins-1 cells cultured in vitro. Methods Ins-1 cells were cultured in vitro together with angiotensin Ⅱ , MTT test was used to menstruate cell proliferation rate and flow cytometry was applied to confirm the rate of early-apoptosis of Ins-1 cells. Results After being cultured with angiotensin Ⅱ for 12 hours, Ins-1 cells showed an obviously reduced proliferation rate and an obviously increased apoptosis rate, and these changes were connected with the different concentrations of angiotensin. Conclusion Angiotensin Ⅱ can induce apoptosis and restrain the proliferation of Ins-1 cells in the specific time and concentration.
